I read somewhere that after the two Na'vi mate there is a bond between them that can actually be seen door other Na'vi. This is how he knew they mated. I cant remember where I read this unfortunately but i will look to get u a link.

Well actually, Neytiri defended Jake after Tsu'Tey attacked him [because he suspected they'd mated]. So I believe it was because he saw Jeytiri holding hands that he figured they were together.
The male and female Na'vi will connect Tsahaylu to create an emotional bond that lasts a lifetime. Once Tsahaylu is made between the couple, the ultimate in intimacy,this causes the somewhat unwillful sharing of the couple's good memories, and is a sign of Eywa's acceptance.
Dudes it's just of those walks of shame ya know. They went into the forest together, met Eywa together *cough* and came out of the forest together. Lol.
Tsu'tey knew they mated for several reasons. 1. When they were flying Jake and Neytiri were flirting with each other sending gusts of wind at each other laughing. 2. They are all connected to Eywa and could feel that connection if looking for it(TsuTey and Neytiri were meant to be mated). 3. The Na'vi have a heightened sense of smell. After they slept together Tsu'tey and his band came across the scene of the bulldozers and if u have subtitles on he inhales deeply as if he is angry but also catches a smell of something else. 4. They walk into hometree holding hands and Neytiri defends Jake when pushed.
